Suarez blocked a goal-bound effort with his hands for which he was shown a straight red card and Ghana's Asamoah Gyan missed the resulting penalty kick in stoppage time of second half extra time.
Uruguay would go on to beat Ghana 4-2 on penalties to advance into the last four, denying the Black Stars the chance to become the first African side to reach the semi-finals.
Twelve years on from that meeting in South Africa, Ghana and Uruguay will meet once again, this time in the group stage in Qatar.
A key subplot to the game will be the memories from the FNB Stadium and what impact it will have this time round, with Ghana looking to seek revenge.
Ahead of the game, Suarez has refused to apologise for the incident and does not understand why it has caused so much uproar.
"I don't apologise for that," he said in a press conference.
"I did the handball, but the Ghana player missed a penalty, not me.
"Maybe I could apologise if I injured the player, but in this situation I was sent off, the ref said penalty – it's not my fault, because I didn't miss the penalty.
"It's not my responsibility to score the penalty."
Any result other than a win for Uruguay will see the South Americans exit the World Cup at the group stage.
