The 25-year-old Brazilian has agreed a five-year contract with the North London club and becomes their fourth summer signing.
Richarlison spent four seasons on the blue half of Merseyside after joining from Watford in the summer of 2018 and scored 53 goals in 152 appearances.
Spurs manager Antonio Conte is keen on bolstering his forward line after they secured Champions League football with a fourth place finish last season.
In a statement, Spurs said: 'We are delighted to announce the signing of Richarlison from Everton, subject to a work permit.
'The Brazilian international has signed a contract with us until 2027.
A statement from Everton read: "Richarlison has completed a transfer from Everton to Tottenham Hotspur for an undisclosed fee.
"The 25-year-old forward departs the Blues after four years at Goodison Park, making 152 appearances and scoring 53 goals.
"Richarlison joined Everton from Watford in July 2018 and, during his time with the Club, made his senior Brazil debut, going on to earn 36 caps for the five-time world champions.
"Everyone at Everton would like to thank Richarlison for his service and commitment during his time with the Club."
Richarlison joins an impressive Spurs forward line that already includes the likes of Harry Kane, Son Heung-Min, Dejan Kulusevski, Lucas Moura and Steven Bergwijn.
Tottenham have already signed Ivan Perisic on a free transfer from Inter Milan this summer as well as goalkeeper Fraser Forster from Southampton and Mali midfielder Yves Bissouma from Brighton and Hove Albion for £30m.
