The want away striker was said to undergo Coronavirus testing on Monday and start training on Tuesday but Kane is yet to appear at the training ground.
After leading England to the Euro 2020 final, losing to Italy on penalties, the England International has enjoyed a three-week break.
Tottenham Hotspurs are yet to disclose the reason to Kane's absence for training.
A move to Manchester City failed to happen before the Euro 2020 tournament but City still remain intact to bring the last season's Premier League top scorer to Etihad as they have also made a £100 million bid for Aston Villa forward Jack Grealish.
Kane who has scored 221 goals is the second all-time top scorer for Spurs and is behind Jimmy Greaves, with 266 times for the club between 1961 and 1970.
Despite pushing for a move away from the Tottenham Hotspurs Stadium, new manager Nuno Espirito Santo is ever ready to start working with the three time Premier League top scorer.
Speaking at his first news conference after being appointed as the new gaffer, Nuno said last month:
"Harry is our player. Period. No need to talk about anything else, I am looking forward to him joining the group.
"Harry is one of the best players in the world. That is all I need to say."
Kane registered 23 goals in thirty-five games with fourteen assists for Spurs in the Premier League last season.
Kane's current contract will keep him at Spurs until 2024 after he signed a six-year deal in 2018.
